fa اثر یک دوره تمرین شنا همراه با مصرف هسپریدین بر بیان ژن‌های مرتبط با بقاء نورون‌های دپامینرژیک در مدل حیوانی پارکینسون Effect of Swimming Training with Hesperidin on Gene Expression Related Dopaminergic Neuron Survival in an Animal Model of Parkinson's Disease فیزیولوژی ورزش Exercise Physiology پژوهشي Original article <div style="text-align: justify;">مقدمه: بیماری پارکینسون با از دست رفتن نورون&rlm;‌‌های دوپامینرژیک در جسم سیاه همراه است. ممکن است فعالیت ورزشی در کنار مصرف آنتی&rlm;‌‌اکسیدان&rlm;‌ها در بقاء این نورون&rlm;‌ها نقش داشته باشد. هدف مطالعه حاضر بررسی اثر یک دوره تمرین شنا همراه با مصرف هسپریدین بر بیان ژن‌&rlm;های مرتبط با بقاء نورون&rlm;های دپامینرژیک در مدل حیوانی پارکینسون بود.</div> <div style="text-align: justify;">روش بررسی: در این مطالعه بنیادی و تجربی، 25 سرموش نر صحرایی نژاد ویستار 8 تا 10 هفته‌ای به‌طور تصادفی در پنج گروه (1) سالم، (2) بیمار، (3) هسپریدین، (4) شنا و (5) شنا+ هسپریدین قرار گرفتند. موش&rlm;‌های گروه‌&rlm;های 4 و 5 به مدت شش هفته تمرین شنا انجام دادند. نمونه&rlm;‌های گروه‌&rlm;های 3 و 5 نیز در این مدت روزانه 500 میلی‌گرم به ازای هر کیلو وزن بدن مکمل هسپریدین دریافت کردند. 48 ساعت پس از آخرین جلسه تمرینی، بیان ژن DJ-1 و LRRK2 در بافت هیپوکامپ با روش Real-time PCR اندازه‌گیری شد. داده&rlm;‌ها با استفاده از تحلیل واریانس یک راهه و آزمون تعقیبی بونفرونی در سطح معنی‌داری 0/05>P و نرم‌افزارversion 16  SPSS مورد تجزیه و تحلیل قرار گرفتند.</div> <div style="text-align: justify;">نتایج: نتایج نشان داد با القاء پارکینسون، بیان ژن DJ-1 نسبت به گروه سالم کاهش در حالی‌که بیان ژن LRRK2 افزایش معنی‌‌داری پیدا کرد (0/05>P). نتایج نشان داد بیان ژن DJ-1 در گروه شنا، هسپریدین و شنا + هسپریدین به‌طور معنی‌داری بیشتر و مقادیر LRRK2 به‌طور معنی&rlm;داری کمتر از گروه بیمار بود (0/001>P). بیان ژن DJ-1 در گروه&rlm;‌های شنا و شنا + هسپریدین به‌طور معنی&rlm;‌داری بیشتر و بیان ژن LRRK2 به‌طور معنی&rlm;‌داری کمتر از گروه هسپریدین بود. علاوه بر این بیان DJ-1 در گروه شنا + هسپریدین به طور معنی&rlm;‌داری بیشتر از گروه شنا بود (0/001>P). </div> <div style="text-align: justify;">نتیجه‌گیری: به‌طور کلی به نظر می‌&rlm;رسد تمرین شنا همراه با مصرف هسپریدین به واسطه افزایش بیان ژن DJ-1 و کاهش بیان ژن LRRK2 در حفظ و بقاء نورون‌های دوپامینرژیک در نمونه‌&rlm;های مبتلا به پارکینسون مؤثر است.</div> <div style="text-align: justify;">Introduction: Parkinson's disease (PD) is a neurodegenerative disorder characterized by the progressive loss of dopaminergic neurons in the substantia nigra. Antioxidant consumption and exercise may be important for these neurons’ survival. The current study looked at how swimming training with hesperidin affected the survival of dopaminergic neurons linked to gene expression in an animal model of Parkinson's disease. Methods: In this experimental study, 25 male Wistar rats aged 8-10 weeks were divided into five groups, including: (1) healthy control, (2) PD (3) hesperidin (4) swimming and (5) swimming + hesperidin. The rats in groups 4 and 5 participated in swimming exercises for six weeks, while the rats in groups 3 and 5 received 500 mg/kg body weight of hesperidin throughout the intervention protocol. Two days following the final intervention session the expressions of the DJ-1 and LRRK2 genes were measured using the Real-time PCR method. The data were analyzed using one-way ANOVA, and Bonferroni post hoc tests were run using SPSS Software version 16 at a significance level of P <0.05. Results: Induction of Parkinson’s disease with reserpine resulted in a notable decrease in DJ-1 gene expression, and an increase in LRRK2 gene expression (P< 0.05). The results showed that DJ-1 gene expression was appreciably elevated in the swimming, hesperidin, and swimming + hesperidin groups, while LRRK2 gene expression was significantly lower compared to the PD group (P < 0.001). The level of DJ-1expression was significantly higher in the swimming and swimming + hesperidin groups (P < 0.001), whereas LRRK2 expression was significantly reduced in these groups compared to the hesperidin-only group. Furthermore, the expression of DJ-1 in the swimming + hesperidin group was significantly higher than that observed in the swimming-only group (P < 0.001).
