fa اثر هشت هفته مکمل دهی ویتامین C و ویتامین E بر پروتئین CP و بیان ژن HSP70 در بافت مغز موش های صحرایی مبتلا به آلزایمر Impact of Eight Weeks of Supplementation with Vitamins C and E on the Expression of CP Protein and the HSP70 Gene in the Brain Tissue of Rats with Alzheimer’s Disease بیوشیمی Biochemistry پژوهشي Original article <div style="text-align: justify;">مقدمه: تفاوت بین مکمل‌های آنتی‌اکسیدانی بر سلامت بیماران مبتلا‌به آلزایمر هنوز به خوبی مشخص نشده‌است. با توجه به اینکه تاکنون مطالعهای به ارزیابی اثربخشی ویتامین C و E بر پروتئین کربونیل (CP) و بیان ژن شوک حرارتی 70 (HSP70) در موشهای آلزایمری نپرداخته است، مطالعه حاضر با هدف بررسی اثر هشت هفته مکمل‌دهی ویتامین C و ویتامین E بر پروتئین CP و بیان HSP70 در بافت مغز موش‌های صحرایی آلزایمری صورت گرفت. روش بررسی: در این مطالعه تجربی تعداد 28 سر موش صحرایی آلزایمری شده با تزریق 10 میلی‌گرم بر کیلوگرم تریمتیلتین به گروه‌های (1) کنترل بیمار، (2) شم (حلال ویتامین E)، (3) ویتامین C و (4) ویتامین E (VE) تقسیم شدند. هم‌چنین تعداد هفت سر موش صحرایی سالم برای بررسی اثرات القا بیماری بر متغیر‌های تحقیق در گروه کنترل سالم قرار گرفتند. مکمل ویتامین C به مدت هشت هفته روزانه 4 میلی‌گرم بر کیلوگرم در روز و مکمل ویتامین E روزانه 30 میلی‌گرم بر کیلوگرم به نمونهها خورانده شد. گروهها از نظر مقادیر CP و بیان ژن HSP70 پس از مکمل‌دهی، مورد بررسی قرار گرفتند. سپس، دادهها توسط نرم‌افزار آماریversion 16  SPSS مورد تجزیه و تحلیل آماری قرار گرفتند. جهت بررسی نرمالیتی توزیع یافتهها از آزمون شاپیرو-ویلک استفاده شد. اثر مکمل ویتامین E و ویتامین C با استفاده از آنالیز وارایانس یک راهه بررسی شد. هم‌چنین، برای بررسی نوع اثر متغیرها از آزمون تعقیبی توکی استفاده شد. سطح معنیداری برای تمام تجزیه و تحلیل‌ها 0/05 در نظر گرفته شد. نتایج: مقادیر پروتئین کربونیل در گروه‌های ویتامین C و ویتامین E به‌طور معنی‌داری کمتر از گروه کنترل بیمار بود (0/05‌>P). هم‌چنین، مقادیر HSP70 در گروه‌های ویتامین C و E به‌طور معنیداری بیشتر از گروه کنترل بیمار بود (0/05‌>P). تفاوتی بین گروه مکمل ویتامین C و E مشاهده نشد (0/05 <P). نتیجه‌گیری: به‌طور خلاصه یافتههای به‌دست آمده از مطالعه حاضر حاکی از آن بود که هر دو ویتامین C و ویتامین E دارای اثرات یکسان آنتی‌اکسیدانی و محافظت سلولی در بافت مغز موشهای مبتلا‌به آلزایمر میباشند. با توجه به تاثیر ویتامین C و E بر کاهش پروتئین کربونیل و افزایش HSP70 در بافت هیپوکامپ موش‌های صحرایی، پیشنهاد میگردد در مطالعات بعدی اثر این ویتامینها بر دیگر اختلالات نورودژنراتیو مورد بررسی قرار گیرد.</div> <div style="text-align: justify;">Introduction: Impact of antioxidant supplements on the health of patients with Alzheimer’s disease has not yet been clearly defined. Given that no study has been conducted so far to evaluate the effectiveness of vitamins C and E on the expression of Carbonyl Protein (CP) and Heat Shock 70 (HSP70) genes in rats with Alzheimer’s, this study aims to investigate the effect of an eight-week supplementation of vitamins C and E on the expression of CP and HSP70 in the brain tissue of rats with Alzheimer’s disease. Methods: In this experimental study, 28 rats with Alzheimer disease were injected with 10 mg/kg of trimethyltin and divided into four groups: patient control, sham (soluble vitamin E), Vitamin C, and Vitamin E (VE). Seven healthy rats were included in the control group to investigate the effects of disease induction on the variables under study. They were administered a daily supplement of 4 mg/kg of vitamin C and 30 mg/kg of vitamin E for eight weeks. After supplementation period, the groups were compared in terms of CP values and HSP70 gene expression. Data were analyzed by SPSS16 statistical software. Shapiro-Wilk test was used to check normality of the finding distributions. Effect of vitamin E and vitamin C supplementation was evaluated using one-way analysis of variance. Tukey's post hoc test was used to check the type of variable effects. A significant level of 0.05 was considered for all analyses. Results: Levels of carbonyl protein in both the vitamin C and vitamin E groups were significantly lower than those in the patient control group (P < 0.05). HSP70 levels in the vitamin C and E groups were significantly higher than those in the patient control group (P < 0.05). No significant difference was observed between vitamin C and E supplement groups (P > 0.05). Conclusion: Findings of this study suggest that both vitamin C and E exhibit similar antioxidant and cellular protective effects in the brain tissue of mice with Alzheimer’s disease.
