fa بررسی شیوع و تعیین ژنوتایپ ویروس هپاتیت دلتا در بیماران HBs Ag مثبت مراجعه‌کننده به کلینیک تخصصی بعثت کرمان طی سال‌های 92-1391 بیماری های عفونی Infectious Diseases پژوهشي Original article <p dir="RTL"><strong>مقدمه: </strong>عامل هپاتیت دلتا، ویروسی با میانگین قطر 35 نانومتر است که جهت تکثیر و تجمع به آنتی‌ژن سطحی ویروس هپاتیت <span dir="LTR">B</span> وابسته است. عفونت با عامل دلتا می‌تواند هم‌زمان با عفونت ویروس هپاتیت <span dir="LTR">B</span> رخ دهد. در این‌صورت معمولاً سبب هپاتیت حاد می‌گردد و یا به شکل عفونت ثانویه در بیماران مبتلا به ویروس هپاتیت <span dir="LTR">B</span> بروز می‌نماید. ویروس هپاتیت دلتا دارای ژنوتیپ‌های مختلفی بر اساس آنالیز توالی ژنوم است که تظاهرات بالینی و گستره جغرافیایی متنوعی را از خود نشان می‌دهند. هدف از این مطالعه، تعیین شیوع و ژنوتیپ ویروس هپاتیت دلتا در بیماران <span dir="LTR">HBsAg</span> مثبت است.</p> <p dir="RTL"><strong>روش بررسی:</strong> این مطالعه از نوع بررسی مقطعی و توصیفی بوده و بر روی 400 نفر بیمار <span dir="LTR">HBsAg</span> مثبت انجام پذیرفت. تعیین ژنوتایپ با استفاده از واکنش زنجیره پلیمراز وهضم آنزیمی با روش<span dir="LTR">(Restriction Fragment Length Polymorphism) RFLP</span> انجام شد. در آخر نیز تعیین سکانس نمونه‌ها برای تائیدنتایج حاصل از<span dir="LTR">RFLP</span> انجام شد.</p> <p dir="RTL"><strong>نتایج: </strong>از بین 400 بیماری که در این تحقیق بررسی شد، 67 مورد(75/16%) دارای آنتی‌بادی برضد <span dir="LTR">HDV</span> بودند که از میان آنها<br> 7 مورد(75/1%) <span dir="LTR">HDV RNA</span> در نمونه سرمی تشخیص داده شد. با بررسی توالی‌ها توسط آنزیم‌های برشی و آنالیز سکانس این 7 نمونه، ژنوتایپ همه از نوع I تعیین گردید.</p> <p dir="RTL"><strong>نتیجه‌گیری: </strong>آنالیز سکانس ناحیه <span dir="LTR">C</span>-ترمینال ویروس هپاتیت دلتا و مقایسه آن با ایزوله‌های سایر کشورها، نشان از یکسان بودن ژنوتایپ ویروس با کشورهای همجواری چون ترکیه، مصر، اردن و پاکستان دارد.احتمالا این یافته‌ها می‌تواند حاکی از منشأ یکسان ویروس دلتا در منطقه خاورمیانه داشته باشد.</p> <p><strong><em>Introduction:</em></strong> Hepatitis D virus (HDV) is a 35 nm in diameter agent that depends on hepatitis B surface antigen in order to proliferate and accumulate. Infection with delta agent can occur spontaneously with hepatitis B virus infection and it cause acute hepatitis or develop as secondary infection in the patients with hepatitis B. Hepatitis D virus (HDV) has several genotypes based on genome sequence analysis, which shows a variety of clinical protests and geographical scope. The aim of this study was to determine the prevalence of hepatitis delta virus genotype in patients with positive HBs Ag.</p> <p><strong><em>Methods:</em></strong> This cross-sectional and descriptive study was performed on 400 HbsAg-positive patients. Genotype identification was performed using polymerase chain reaction and RT-nested PCR and RFLP (Restriction Fragment Length Polymorphism). Finally sample sequencing was considered to approve RFLP gained results.</p> <p><strong><em>Results: </em></strong>Among 400 patients in this study, 67 participants (16.75 %) were containing anti-HDV antibody, and it was found HDV RNA in just 7 (1.75%) serum samples. By examining the sequences using analysis of these 7 positive HDV RNA samples by restriction enzyme and MEGA4 software showed that all of them have genotype I.</p> <p><strong><em>Conclusion:</em></strong> Sequence analysis of C-terminal region of Hepatitis D virus (HDV) and its comparison with other countries showed that it was the same genotype that present in countries such as Turkey, Egypt, Jordan and Pakistan.
